Millions of us are successfully treated with ciprofloxacin and other fluoroquinolone antibiotics each year. But for some patients the drugs are linked to severe adverse reactions involving terrifying physical and mental health impacts. Andrew Wasley reports Patients who say they’ve suffered severe adverse reactions to a common antibiotic are calling for action to prevent others from experiencing a 'frightening' number of alleged physical and mental side effects. The victims, who say they were poisoned by ciprofloxacin, want more research into the drug’s side effects, greater education of health professionals and clearer warnings for consumers. The calls come as an investigation revealed the antibiotic has been linked to more than forty deaths in the UK in recent years, and been the subject of hundreds of suspected adverse reactions. Ciprofloxacin, part of the fluoroquinolone class of antibiotics, is prescribed to treat a range of medical conditions, including bacterial infections. Fluoroquinolones are also – controversially – used to treat diseases in poultry, pig and cattle farming. If you're sick and you're feeling really, really tired, it may not be just the illness itself that has you dragging. It could be the antibiotics you're taking to cure it that is the problem. Amoxicillin, which is used to treat bacterial infections in various parts of the body, can cause tiredness or weakness as a side effect, and patients should alert their doctor if that happens [source: Mayo Clinic, Healthline]. The antibiotic Azithromycin also can cause extreme tiredness as a side effect, and again, patients should seek medical assistance if they experience it [source: Medline Plus]. Patients taking ciprofloxacin, an antibiotic that became famous as a cure for anthrax poisoning, also should get medical help if they experience unusual tiredness [source: Medline Plus]. Unfortunately, while researchers have known for a long time that some patients who take antibiotics experience debilitating fatigue as a result, they haven't yet figured out why. One problem is that the result has been difficult to duplicate in experiments.

These tears, called aortic dissections, or ruptures of an aortic aneurysm can lead to dangerous bleeding or even death. They can occur with fluoroquinolones for systemic use given by mouth or through an injection. BACKGROUND: Fluoroquinolone antibiotics are approved to treat certain bacterial infections and have been used for more than 30 years. They work by killing or stopping the growth of bacteria that can cause illness. Without treatment, some infections can spread and lead to serious health problems (see List of Currently Available FDA-Approved Systemic Fluoroquinolones, available at RECOMMENDATION: Healthcare professionals should: Taking ciprofloxacin increases the risk that you will develop tendinitis (swelling of a fibrous tissue that connects a bone to a muscle) or have a tendon rupture (tearing of a fibrous tissue that connects a bone to a muscle) during your treatment or for up to several months afterward. The FDA has issued new warnings for a group of antibiotics called fluoroquinolones after a safety review confirmed what many patients have long-considered to be disabling side effects including irregular heartbeat, depression, nerve damage, ruptured tendons, and seizures. The new FDA ruling calls for restricted use of fluoroquinolones including: ciprofloxacin (Cipro), levofloxacin (Levaquin), moxifloxacin (Avelox), ofloxacin (Floxin), and gemifloxacin (Factive). D., an infectious disease physician at Brigham and Women’s Hospital in Boston, Massachusetts, and a member of the FDA panel, “this class of antibiotics should be reserved for a second-line or even third-line treatment after other antibiotics have failed.” Side Effects a Result of Mitochondrial Toxicity Though the FDA is now requiring stricter warnings on these drug labels, these concerns are not new. An FDA expert advisory panel determined that the “potentially disabling and permanent” side effects caused by fluoroquinolones outweigh their benefit in treating ailments such as bronchitis, sinus infections, and urinary tract infections. Several studies, including an earlier FDA review in 2013, previously pointed to mitochondrial damage as an underlying mechanism for several of these side effects. In a 2013 Pharmacovigilance Review, the FDA reported results from a study evaluating oxidative stress in patients taking ciprofloxacin, levofloxacin, and gatifloxacin for complicated urinary tract infections. The study showed “increases in lipid peroxides can quickly overwhelm cellular antioxidants, leading to impairment of cell integrity and cell death.” In vitro studies of ciprofloxacin have demonstrated a loss of mitochondrial DNA (mt DNA), resulting in a decrease of mitochondrial respiration and retarded cell growth. Following this review, the FDA issued a Drug Safety Communication on August 15, 2013 notifying the public that it was requiring the drug labels and Medication Guides for all fluoroquinolone antibiotics to be updated to better describe the serious side effect of peripheral neuropathy. The notice stated, “serious nerve damage potentially caused by fluoroquinolones may occur soon after these drugs are taken and may be permanent.” This was not the first warning about the detrimental effects of fluoroquinolones.
